Split PCR Primers set

The Split PCR Primers Set is designed for the rapid construction of expression templates using the Split Primer PCR method.
This method uses a two-step PCR process to add an SP6 promoter, the E01 translational enhancer sequence, and a 3′ UTR to the target protein-coding sequence, enabling high-throughput preparation of linear DNA expression templates suitable for wheat germ cell-free protein synthesis.
The set also includes primers for constructing templates to produce His-tagged fusion proteins for purification.

The Split Primer PCR method enables rapid construction of expression templates, allowing the synthesis and His-tag purification of up to 96 different proteins at a reaction volume of 227 µL per protein.

WEPRO7240H has been depleted of endogenous histidine-rich proteins derived from wheat germ, thereby improving purification performance. Compared with the WEPRO7240 Core Kit, it enables the production of His-tagged proteins with higher purity.
